Agar, Bacteriological No. 1 is a solidifying agent for use in preparing microbiological culture media in a laboratory setting. Agar, Bacteriological No. 1 is not intended for use in the diagnosis of disease or other conditions in humans. Agar, Bacteriological No. 1 conforms with the testing specified in the USP/EP Pharmacopeia. Agar is a phycocolloid extracted from a group of red-purple algae, usually Gelidium spp. Agar was first suggested for microbiological purposes in 1881 by Fannie Hesse. By the early 1900’s, agar became the gelling agent of choice over gelatin because agar remains firm at growth temperature for many pathogens and agar is generally resistant to a breakdown by bacterial enzymes. The use of agar in microbiological media significantly contributed to the advance of microbiology, paving the way to study pure cultures. Agar is a gel at room temperature, remaining firm at temperatures as high as 65°C. Agar melts at approximately 85 - 91°C, a different temperature from solidification at 34 - 36°C. This property is known as hysteresis. Agar is generally resistant to shear forces; however, different agar may have different gel strengths or degrees of stiffness. Specifications for Agar, Bacteriological No. 1 include good clarity, controlled gelation temperature, controlled melting temperature, good diffusion characteristics, absence of toxic bacterial inhibitors, and relative absence metabolically useful minerals and compounds. Agar, Bacteriological No. 1 is recommended for clinical applications, auxotrophic studies, bacterial and yeast transformation studies, and bacterial molecular genetics applications.

Brilliant Green Agar w/ Sulfapyridine is used for the selective enrichment of Salmonella spp. in a laboratory setting. Brilliant Green Agar w/ Sulfapyridine is not intended for use in the diagnosis of disease or other conditions in humans. Brilliant Green Agar was first described by Kristensen et al and later modified by Kauffmann. The outstanding selectivity of this medium permits the use of moderately heavy inocula evenly distributed over the surface. The addition of sulfonamides into Brilliant Green Agar further inhibits Escherichia coli and Proteus spp. Osborne and Stokes used 0.1% Sodium Sulfapyridine to enhance the recovery of Salmonella from whole egg and egg yolk.

Columbia CNA Agar is used with blood for the selective isolation of Gram-positive cocci in a laboratory setting. Columbia CNA Agar is not intended for use in the diagnosis of disease or other conditions in humans. Ellner et al. described Columbia CNA Agar as a variation of Columbia Blood Agar Base that is selective for Gram-positive cocci. Colistin and Nalidixic Acid are added to the formula, selecting for Gram-positive organisms and fungi by suppressing Gram-negative bacteria.

Campylobacter Blood-Free Selective Medium (Modified Charcoal Cefoperazone Deoycholate Agar or mCCDA) is used for the cultivation of campylobacters, and is not intended for use in the diagnosis of disease or other conditions in humans. A blood free medium which will support the growth of most enteric campylobacters. The selective supplement X112 (X212) makes the medium selective for C. jejuni. and C. coli when incubated at 37°C. With this product incubation at 42°C is no longer necessary and higher recovery rates have been reported at 37°C than at 42°C. The supplement X112 (X212) consists of cefoperazone and amphotericin and is superior to the selective combination of Skirrow, Butzler and Blazer-Wang all of which contain antibiotics shown to be inhibitors to C. coli. The colonial morphologies of Campylobacter spp. on this medium are distinctive.
