Rappaport Vassiliadis R10 Broth is used for the selective enrichment of Salmonella spp. from foods in a laboratory setting. Rappaport Vassiliadis R10 Broth is not intended for use in the diagnosis of disease or other conditions in humans. Rappaport et al formulated an enrichment medium for Salmonella spp. that was modified by Vassiliadis et al. The Rappaport formulation, designated R 25/37°C, recommended incubation at 37°C. The Vassiliadis modification, designated R 10/43°C, had a reduced level of Malachite Green and recommended incubation at 43°C. Peterz later showed that incubation at 41.5 ± 0.5°C for 24 hours improved recovery of Salmonella spp. Rappaport-Vassiliadis R10 Broth is a selective enrichment medium that is used following pre-enrichment of the specimen. It has gained approval for use in analyzing milk and milk products, raw flesh foods, highly contaminated foods, and animal feeds. This medium selectively enriches for Salmonella spp. because bacteria, including other intestinal bacteria, are typically susceptible to or inhibited by Malachite Green, high osmotic pressure and/or low pH. S. typhi and S. choleraesuis are sensitive to Malachite Green and may be inhibited.

YM Broth is used for the cultivation of fungi in a laboratory setting. YM Broth is not intended for use in the diagnosis of disease or other conditions in humans. YM Broth is prepared according to the formulation published by Wickerham. Wickerham suggested that YM Broth, acidified to pH 3.0 – 4.0, be used as an enrichment medium for yeasts from populations also containing bacteria and molds. To favor isolation of fermentative species, add a layer of sterile paraffin oil 1 cm deep on the surface of the inoculated broth. Cultures should be incubated until growth appears, and then streaked onto YM Agar to obtain isolated yeast colonies. To isolate fermentative and oxidative strains, place acidified inoculated YM Broth on a rotary shaker for 1 to 2 days. This favors yeast recovery while preventing sporulation of molds. Media selectivity may be enhanced through acidification or addition of selective agents. YM Broth may be acidified prior to sterilization.

Listeria monocytogenes, described first in 1926 by Murray, Webb, and Swann, is an extensive problem in public health and food industries. Evidence from outbreaks of listeriosis has indicated that the principle route of transmission is via consumption of foodstuffs contaminated with Listeria monocytogenes. Implicated vehicles of transmission include turkey, frankfurters, coleslaw, pasteurized milk, Mexican style cheese, and pate. Listeria spp. are ubiquitous in nature, being present in a wide range of unprocessed foods as well as in soil, sewage, and river water.
Buffered Listeria Enrichment Broth, a modification of the formula by Lovett et al., was developed after subsequent work concluded that enrichment properties can be improved by increasing the buffering capacity of the medium with the addition of Disodium Phosphate. Listeria spp. grow over a pH range of 5.0 - 9.6, and survive in food products with pH levels outside these parameters. Listeria spp. are microaerophilic, Gram-positive, asporogenous, non-encapsulated, non-branching, short, motile rods. Motility is pronounced at 20°C. Identification of Listeria is based on successful isolation of the organism, biochemical characterization, and serological confirmation.

In 1948, Weber and Black described the value of a highly nutritional solid medium containing neutralizing agents for quaternary ammonium compounds in sanitisers. The addition of Lecithin and Polysorbate 80 to Tryptone Glucose Extract (TGE) Agar resulted in a medium that effectively neutralizes quaternary ammonium compounds in testing of germicidal activity. Total neutralization of disinfectants is critical. Disinfectant residues can result in a false negative (no-growth) test.
Letheen Broth Base Modified without Tween is based on the formula described in FDA Bacteriological Analytical Manual, and a modification of Letheen Broth Base. Letheen Broth Base Modified is recommended by the FDA for use in the microbiological testing of cosmetics.

Lauryl Sulfate Broth is used for the detection of coliform bacteria in water and wastewater in a laboratory setting. Lauryl Sulfate Broth is not intended for use in the diagnosis of disease or other conditions in humans.
The coliform group of bacteria includes aerobic and facultative anaerobic, Gram-negative, non-sporeforming bacilli that ferment lactose and form acid and gas at 35°C within 48 hours. Members of the Enterobacteriacae comprise the majority of this group, but organisms such as Aeromonas spp. may also be included. Procedures to detect and confirm coliforms are used in testing water, foods, dairy products, and other materials.
Lauryl Sulfate Broth, also referred to as Lauryl Tryptose Broth, is prepared according to the formula of Mallmann and Darby. During their investigation, Sodium Lauryl Sulfate produced the best results for inhibition of organisms other than coliforms. Lauryl Sulfate Broth, abbreviated as LSB, is used in the presumptive phase of the Standard Total Coliform Fermentation Technique in the examination of water, and coliform detection of foods.

Salmonella organisms may be present but injured in food-processing procedures, which include exposure to low temperatures, sub-marginal heat, drying, radiation, preservative, and sanitisers. Numerous reported cases of Salmonella enteritidis (SE) infections are associated with contaminated shell eggs. SE infections can be very serious and symptoms can include diarrhea, fever, abdominal cramps, headache, nausea and vomiting. Eggs can become contaminated on the farm because a laying hen can become infected with SE and pass the bacteria into the egg before it is laid. If the egg is not refrigerated, the bacteria can grow inside the uncracked, whole egg. Tetrathionate Broth Base, Hajna was developed by Hajna and Damon.

Lauryl Tryptose (LST) Broth is used for the detection of coliform bacteria in water and wastewater. Lauryl Tryptose Broth is not intended for use in the diagnosis of disease or other conditions in humans. Lauryl Tryptose Broth is a selective medium for the detection of coliforms in water, dairy products and other foods. The American Public Health Authority (APHA) recommend Lauryl Tryptose Broth for the Most Probable Number Presumptive Test of coliforms in waters, effluent or sewage and as a confirmation test of lactose fermentation with gas production from milk samples and for the detection of coliforms in foods. Lauryl Tryptose Broth is prepared according to the formulation of Mallmann and Darby. Mallmann and Darby showed that tryptose at a concentration of 2% increased the early logarithmic growth phase when compared to meat peptone.
These researchers added phosphate buffers and sodium chloride, which improved gas production by "slow lactose fermenting" organisms. Sodium lauryl sulfate was incorporated as a selective agent for the inhibition of non-coliform organisms. This medium can also be used with the addition of MUG (4- methylumbelliferyl-β-D-glucuronide) according to the ISO Standard 11866-2 to give enhanced detection of Escherichia coli.

EC Broth is made selective for coliforms by the inclusion of Bile Salts in the dehydrated medium. The selective nature of this medium ensures that the growth of non-coliform bacteria is minimized. The medium is buffered by the addition of potassium phosphates and osmotically balanced by sodium chloride. The medium is used at 37°C for coliform organisms and 44.5°C is recommended for the isolation E. coli.

Modified Tryptone-Soy Broth has emerged as the medium of choice for the enrichment of E. coli O157:H7 in red meats and is incorporated into ISO/TS 13136:2012. This medium is not intended for use in the diagnosis of disease or other conditions in humans. Concern regarding E. coli O157:H7 has grown due to the severity of the disease syndromes caused, and the increase in foodborne infection, so too has the need to optimize methods for its efficient isolation. Symptoms start with severe stomach cramps and watery, bloody diarrhea, and a percentage of individuals infected will develop Haemolytic Uraemic Syndrome (HUS) leading to acute renal failure. In a comparison of four different selective broth media, MTSB was the most productive and selective for the isolation of E. coli O157:H7. mTSB is made selective for O157:H7 by including bile salts in the dehydrated medium, and the addition of novobiocin supplement (X150).

M-Green Yeast and Fungi Broth is a relatively more complex formula compared to other media used for isolation of fungi and yeast. This formulation is rich in nutrients which allows for excellent fungal growth. Bacterial growth is inhibited by an acid pH.
Fungi have been found in potable water and on the inner surface of distribution system pipes. They can survive water treatment or they enter the system after treatment and remain viable.

Lactobacilli MRS Broth is used for the cultivation of lactobacilli and is not intended for use in the diagnosis of disease or other conditions in humans.
Lactobacilli MRS broth was originally developed in 1960 by de Man, Rogosa & Sharpe, the medium can be used for confirmatory tests on organisms isolated on MRS Agar. The medium can also be used for enumeration by the Miles and Misra technique.
Nutrition is provided by a mixture of carefully selected peptones, glucose, beef and yeast extracts while Tween® 80, magnesium and manganese sulfates act as growth stimulants. Selectivity against streptococci & molds is provided by ammonium citrate and sodium acetate.
Occasionally, sterilization of this medium at 121°C for 15 minutes, in some autoclaves, may cause the pH to fall outside of the specified pH limits 6.4 +/- 0.2. In these rare cases, adjustment of the medium using acetic acid or sodium hydroxide is recommended.

Tryptone Soy Broth (TSB) is a general-purpose broth which will support the growth of a wide range of micro-organisms and is not intended for use in the diagnosis of disease or other conditions in humans. Tryptic Soy Broth, a general purpose medium, is commonly referred to as Casein Soya Bean or Soybean- Casein Digest Broth, and abbreviated as TSB. This medium was originally developed for use without blood in determining the effectiveness of sulfonamides against pneumococci and other organisms. Clostridia and non-sporulating anaerobes grow luxuriantly in this broth when incubated under anaerobic conditions. TSB is recommended for testing bacterial contaminants in cosmetics and complies with established standards in the food industry. Enzymatic Digest of Casein and Enzymatic Digest of Soybean Meal are nitrogen sources in TSB. Dextrose is the carbon energy source that facilitates organism growth. Sodium Chloride maintains osmotic balance; Dipotassium Phosphate is a buffering agent.
