The media that Gem Scientific use is designed to not only provide you with the most accurate results but visually distinctive dependent on the product type and the level of bacteria found at the original testing location

Within each box of Gem Scientific dip slides is a handy comparison chart to demonstrate what level of contamination the tested area has. This is measured by the coverage of the media by the bacteria recovered from the sample or area tested. Regular testing is recommended to ensure your regimes are being maintained and bacteria levels are as low as possible.

Storage

Store in a cool dry place (Optimum 8-15oC) and use before the expiry date on the box. Do not use slides if there is any sign of growth before opening. Slides remain usable while the agar is smooth and attached to the slide. A small amount of water may collect at the bottom of the tube. This is condensate from the agar and does not affect performance, provided the agar still fills the tray.

Disposal

Used slides should be incinerated, autoclaved or soaked in disinfectant for several hours before disposal.

YM Broth is used for the cultivation of fungi in a laboratory setting. YM Broth is not intended for use in the diagnosis of disease or other conditions in humans. YM Broth is prepared according to the formulation published by Wickerham. Wickerham suggested that YM Broth, acidified to pH 3.0 – 4.0, be used as an enrichment medium for yeasts from populations also containing bacteria and molds. To favor isolation of fermentative species, add a layer of sterile paraffin oil 1 cm deep on the surface of the inoculated broth. Cultures should be incubated until growth appears, and then streaked onto YM Agar to obtain isolated yeast colonies. To isolate fermentative and oxidative strains, place acidified inoculated YM Broth on a rotary shaker for 1 to 2 days. This favors yeast recovery while preventing sporulation of molds. Media selectivity may be enhanced through acidification or addition of selective agents. YM Broth may be acidified prior to sterilization.

Lactobacilli MRS Broth is used for the cultivation of lactobacilli and is not intended for use in the diagnosis of disease or other conditions in humans. Lactobacilli MRS broth was originally developed in 1960 by de Man, Rogosa & Sharpe, the medium can be used for confirmatory tests on organisms isolated on MRS Agar. The medium can also be used for enumeration by the Miles and Misra technique. Nutrition is provided by a mixture of carefully selected peptones, glucose, beef and yeast extracts while Tween® 80, magnesium and manganese sulfates act as growth stimulants. Selectivity against streptococci & molds is provided by ammonium citrate and sodium acetate. Occasionally, sterilization of this medium at 121°C for 15 minutes, in some autoclaves, may cause the pH to fall outside of the specified pH limits 6.4 +/- 0.2. In these rare cases, adjustment of the medium using acetic acid or sodium hydroxide is recommended.

Bacillus Cereus Agar Base is used with Polymyxin B and Egg Yolk Emulsion for the isolation and presumptive identification of Bacillus cereus and is not intended for use in the diagnosis of human disease. Holbrook and Anderson described a highly selective and diagnostic medium (PEMBA) for the isolation and determination of Bacillus cereus from food. The medium is formulated to detect small numbers of B. cereus in the presence of large number of contaminants. This medium differentiates B. cereus from other bacteria based on resistance to polymyxin, lack of mannitol fermentation, and presence of lecithinase. B. cereus can be found on vegetables, processed foods, and in nature. B. cereus causes gastrointestinal illness if the organism is allowed to proliferate. Outbreaks of foodborne illness have been associated with boiled and cooked rice, cooked meats, and cooked vegetables.
