During incubation with milk, resazurin sodium salt undergoes reduction (redox) and the colour changes from blue to pink, which indicates highly fluorescent resorufin. then to colourless dihydroresorufin (non fluorescent). The rate of colour change is due to the lowering of the oxidation-reduction potential in the milk, so is an indication of the presence of microbial organisms in the sample.
A standard solution of resazurin is prepared by dissolving one tablet in 50ml of sterile glass-distilled water. One ml of this solution should be added to 10ml of milk for each test. Solutions must be made fresh each day.
