Tryptone Glucose Extract (TGE) Agar is used for the cultivation and enumeration of microorganisms in dairy and bottled water products. Tryptone Glucose Extract (TGE) Agar is not intended for use in the diagnosis of disease or other conditions in humans. In the 1930’s, Bower and Hucker developed a medium for detecting bacteria in milk and other dairy products. Prickett used a glucose agar containing tryptone to study thermophilic bacteria in milk. This medium is known as Yeast Dextrose Agar. In 1948, the American Public Health Association (APHA) adopted Tryptone Glucose Extract Agar for use in testing milk and dairy products. Currently, APHA specifies Tryptone Glucose Extract Agar for the heterotrophic plate count procedure in testing bottled water. Tryptone Glucose Extract Agar is also known as Yeast Dextrose Agar.

Beef Extract Powder is a dehydrated extract of bovine tissue for use in preparing microbiological culture media in a laboratory setting. Beef Extract Powder is not intended for use in the diagnosis of disease or other conditions in humans. Beef Extract Powder is prepared and standardized for use in microbiological culture media, where it is generally used to replace infusion of meat. Culture media containing Beef Extract Powder are recommended for use in bacteriological examination of water, milk, and other materials, where uniform composition of media is important. Beef Extract Powder is relied upon for biochemical studies, particularly fermentation reactions because of its independence from fermentable substances. Several media containing Beef Extract Powder are recommended in standard methods for multiple application.

Listeria Enrichment Broth is used for selective enrichment of Listeria spp and is not intended for use in the diagnosis of disease or other conditions in humans. Listeria monocytogenes, first described in 1926 by Murray, Webb, and Swann, is an extensive problem in public health and food industries. This organism has the ability to cause human illness and death, particularly in immunocompromised individuals and pregnant women. Epidemiological evidence from outbreaks of listeriosis has indicated that the principle route of transmission is via consumption of foodstuffs contaminated with Listeria monocytogenes. Implicated vehicles of transmission include turkey frankfurters, coleslaw, pasteurized milk, Mexican style cheese, and pate. Listeria spp. are ubiquitous in nature, being present in a wide range of unprocessed foods as well as in soil, sewage, and river water. Listeria Enrichment Broth is based on the formula developed by Lovett et al. in which Tryptic Soy Broth is supplemented with Yeast Extract for optimum growth. Listeria spp. grow over a pH range of 5.0 - 9.6, and survive in food products with pH levels outside these parameters. Listeria spp. are microaerophilic, Gram-positive, asporogenous, non-encapsulated, non-branching, short, motile rods. Motility is pronounced at 20°C. Identification of Listeria is based on successful isolation of the organism, biochemical characterization, and serological confirmation.

Yersinia Selective Agar is used with cefsulodin and novobiocin for the isolation of Yersinia enterocolitica in a laboratory setting. Yersinia Selective Agar is not intended for use in the diagnosis of disease or other conditions in humans. This medium is based on the work of Schiemann. It is used for the isolation and enumeration of Yersinia spp. from clinical samples and from food. The selective components are sodium desoxycholate, crystal violet, cefsulodin, irgasan and novobiocin. Yersiniae ferment mannitol with an intense, localized, acid production in the center of the colony which produces a red ‘bull’s eye’ appearance. The ratio of transparent border to red centre varies with serotype and environmental strains may appear rough with an irregular edge. Most other enteric bacteria, if they grow, produce a larger colony with a diffuse pinkish center and opaque outer zone.
