Bacillus Cereus Agar Base is used with Polymyxin B and Egg Yolk Emulsion for the isolation and presumptive identification of Bacillus cereus and is not intended for use in the diagnosis of human disease. Holbrook and Anderson described a highly selective and diagnostic medium (PEMBA) for the isolation and determination of Bacillus cereus from food. The medium is formulated to detect small numbers of B. cereus in the presence of large number of contaminants. This medium differentiates B. cereus from other bacteria based on resistance to polymyxin, lack of mannitol fermentation, and presence of lecithinase. B. cereus can be found on vegetables, processed foods, and in nature. B. cereus causes gastrointestinal illness if the organism is allowed to proliferate. Outbreaks of foodborne illness have been associated with boiled and cooked rice, cooked meats, and cooked vegetables.

A nutrient agar corresponding to the standard formulation for the plate count of micro-organisms in water and dairy products. This medium is also useful for teaching and demonstration purposes using non-fastidious organisms.

Yeast and Mold Agar is recommended for the isolation and maintenance of yeasts and molds. This medium can also be used for the detection of wild yeasts in beer. Yeast and Mold Agar is not intended for use in the diagnosis of disease or other conditions in humans. Yeast and Mold Agar is prepared according to the formulation published by Wickerham and was formulated for the selective isolation of yeasts from mixed cultures containing bacteria and molds. The selectivity of this medium may be enhanced through acidification or through addition of selective agents. Yeast and Mold Agar should be sterilized without pH adjustment and sterile acid added to the medium cooled to 45 - 50°C. Acidified Yeast and Mold Agar should not be heated. Antibiotics may be aseptically added to sterile media. Other fungistatic materials may also be added to Yeast and Mold Agar to eliminate molds and permit enumeration of yeasts to mixed populations.

This medium is not intended for use in the diagnosis of disease or other conditions in humans. The replacement of neutral red used in the original formulation by bromocresol purple makes the color change caused by acid producing organisms easier to read.

Eosin Methylene Blue Agar Levine is used for the isolation and differentiation of Gram-negative enteric bacilli and is not intended for use in the diagnosis of disease or other conditions in humans. This medium was introduced in 1916 by Holt-Harris and Teague to differentiate Escherichia spp. and Aerobacter spp. It was modified by Levine in 1918 who removed sucrose from the formula and increased the lactose content. The distinctive metallic sheen produced by E. coli on this medium is due to acid production resulting in an amide bonding between the eosin and methylene blue, other coliforms do not produce enough acid to cause this reaction. Eosin inhibits most Gram-positive organisms. The prepared medium is sensitive to light.
