LB Agar (Miller), a nutritionally rich medium designed for growth of pure cultures of recombinant strains, is based on the formulation described by Miller. E. coli is grown to late log phase in LB Medium. Some plasmid vectors replicate to a high copy number and do not require selective amplification. Some vectors do not replicate so freely and need to be selectively amplified. Chloramphenicol may be added to inhibit host synthesis and prevent replication of the bacterial chromosome. LB Agar, Miller contains 10 g/L of sodium chloride, different from the levels in Lennox formulations. This allows the researcher to select the optimal salt concentration for a specific strain. The medium may be aseptically supplemented with glucose or antibiotics.

Skim Milk, Agglomerated NFDM is dehydrated skim milk for use in preparing microbiological culture media in a laboratory setting. Skim Milk, Agglomerated NFDM is not intended for use in the diagnosis of disease or other conditions in humans. Skim Milk, Agglomerated NFDM is soluble, spray-dried skim milk. When prepared in a 10% solution, it is equivalent to fresh skim milk. Skim Milk is added to enhance growth of Mycobacterium tuberculosis and Corynebacterium diphtheria in culture media. Skim Milk, Agglomerated NFDM is used for differentiating organisms based on coagulation and proteolysis of casein in foods and dairy products. Indicators of pH, Litmus and Bromocresol Purple, are used with Skim Milk, Agglomerated NFDM to determine acid production. Methylene Blue and Resazurin may be used with Skim Milk, Agglomerated NFDM as oxidation-reduction indicators.

Dextrose Tryptone Agar is used for isolation of mesophilic or thermophilic spoilage microorganisms from food and is not intended for use in the diagnosis of disease or other conditions in humans. Dextrose Tryptone Agar evolved from research by Williams, while studying the cultivation and enumeration of thermophilic bacteria caused by “flat-sour” spoilage of canned foods. In the 1930’s, the National Canners Association specified the use of Dextrose Tryptone Agar for isolating “flat sour” organisms from food products. “Flat sour” spoilage of canned foods is caused by Bacillus coagulans (Bacillus thermoacidurans). Bacterial growth results in a 0.3 – 0.5 drop in pH, while ends of the can remain flat. B. coagulans is a soil microorganism, found in canned tomato products and dairy products. Conditions favorable for organism growth can result in spoilage of food products. Dextrose Tryptone Agar can be used to isolate other food spoilage bacteria including mesophilic, aerobic spore-formers and thermophilic “flat sour” spore-formers such as B. stearothermophilus.

Dermatophyte Test Medium is used for the selective isolation of dermatophytic fungi and is not intended for use in the diagnosis of disease or other conditions in humans. In 1969, Taplin et al. developed this medium for the isolation and recognition of dermatophytic fungi, the causative agent of ringworm from hair, nails, and skin. Dermatophyte Test Medium is preferred for isolation and early recognition of Microsporum, Trichophyton, and Epidermophyton genera because of a distinct color change in the medium. Rapidly-growing species may produce a complete color change in the medium in 3 days. The slower-growing species will change the indicator in longer time periods. Other organisms may grow but can be recognized as non-dermatophytes by lack of a color change. A few organisms, including saprophytes, yeasts, and bacteria are capable of changing the medium from red to yellow, but are easily recognized by their distinctive colonial morphology.
