Thiosulfate Citrate Bile Salts Sucrose (TCBS) Agar is used for the selective isolation of Vibrio cholerae and other enteropathogenic vibrios according to ISO 21872-1:2017 and ISO 21872-2:2007. Thiosulfate Citrate Bile Salts Sucrose (TCBS) Agar is not intended for use in the diagnosis of disease or other conditions in humans. TCBS Agar is also called Vibrio Selective Agar, and prepared according to the formula of Kobayashi et al. This formula is a modification of the selective medium from Nakanishi. All pathogenic Vibrio spp., except Vibrio hollisae, will grow on TCBS Agar. This highly selective agar meets the nutritional requirements of Vibrio spp. Vibrio spp. are able to grow in media containing increased salt concentrations, and some species are halophilic. Infections have been associated with ingestion of contaminated water and consumption of contaminated shellfish or seafood. Vibrio spp. are natural inhabitants of seawater. According to ISO 21872-2, pre-incubation in Alkaline Saline Peptone Water (ASPW) (NCM0175) is required.

EC Broth is used for the detection of coliform bacteria at 37°C and Escherichia coli at 44.5°C. EC Broth is not intended for use in the diagnosis of disease or other conditions in humans. EC Broth ( Escherichia coli Broth) is a selective enrichment broth designed for the isolation of coliforms, including E. coli, from water and food samples. It was the recommended medium of the American Public Health Association (APHA) and the AOAC. EC Broth is made selective for coliforms by the inclusion of Bile Salts in the dehydrated medium. The selective nature of this medium ensures that the growth of non-coliform bacteria is minimized. The medium is buffered by the addition of potassium phosphates and osmotically balanced by sodium chloride. The medium is used at 37°C for coliform organisms and 44.5°C is recommended for the isolation E. coli.

Agar, Bacteriological No. 2 is a solidifying agent for use in preparing microbiological culture media in a laboratory setting. Agar, Bacteriological No. 2 is not intended for use in the diagnosis of disease or other conditions in humans. Agar is a phycocolloid extracted from a group of red-purple algae, usually Gelidium spp. Agar was first suggested for microbiological purposes in 1881 by Fannie Hesse. By the early 1900’s, agar became the gelling agent of choice over gelatin because agar remains firm at growth temperature for many pathogens and agar is generally resistant to a breakdown by bacterial enzymes. The use of agar in microbiological media significantly contributed to the advance of microbiology, paving the way to study pure cultures. Agar is a gel at room temperature, remaining firm at temperatures as high as 65°C. Agar melts at approximately 85 - 91°C, a different temperature from solidification at 34 - 36°C. This property is known as hysteresis. Agar is generally resistant to shear forces; however, different agar may have different gel strengths or degrees of stiffness. Specifications for Agar, Bacteriological No. 2 include good clarity, controlled gelation temperature, controlled melting temperature, good diffusion characteristics, absence of bacterial inhibitors, and the presence of metabolically useful minerals such as calcium and magnesium. Agar, Bacteriological No. 2 is recommended for various applications, auxotrophic studies, bacterial and yeast transformation studies, and bacterial molecular genetics applications.
