Brilliant Green Agar is used for the selective isolation of Salmonella spp. in a laboratory setting. Brilliant Green Agar is not intended for use in the diagnosis of disease or other conditions in humans. Kristensen, Lester, and Jurgens first described the use of Brilliant Green Agar as a primary plating medium for the isolation of Salmonella. The report described the medium as useful for the differentiation of “paratyphoid B” from other intestinal gram-negative bacilli. Kaufmann modified their formula and used Brilliant Green Agar in addition to Tetrathionate Broth for the isolation of Salmonella from stool specimens. The outstanding selectivity of this medium permits use of moderately heavy inocula, which should be evenly distributed over the surface. Brilliant Green Agar is used in the microbial limits test. Brilliant Green Agar supplemented with novobiocin is used in food testing.

Mueller Hinton Agar II is used in antimicrobial susceptibility testing by the disk diffusion method. This formula conforms to Clinical and Laboratory Standard Institute (CLSI), formerly National Committee for Clinical Laboratory Standards (NCCLS). Mueller Hinton Agar II is based on the formula recommended by Mueller and Hinton for the primary isolation of Neisseria species. Mueller and Hinton selected pea meal extract agar as a simple transparent medium containing heat stable ingredients. During their modification, starch replaced the growth-promoting properties of pea extract, acting as a “protective colloid” against toxic substances. Bauer, Kirby, Sherris and Tuck recommended Mueller Hinton Agar for performing antibiotic susceptibility tests using a single disk of high concentration. This un-supplemented medium has been selected by the Clinical and Laboratory Standard Institute (CLSI) for several reasons. This medium is low in sulfonamide, trimethoprim and tetracycline inhibitors, and provides satisfactory growth of most non-fastidious pathogens along with demonstrating batch-to-batch reproducibility. Mueller Hinton Agar is often abbreviated as M-H Agar and complies with requirements of the World Health Organization. Mueller Hinton Agar is specified in FDA Bacteriological Analytical Manual for food testing, and procedures commonly performed on aerobic and facultatively anaerobic bacteria. A variety of supplements can be added to Mueller Hinton Agar, including 5% defibrinated sheep or horse blood, 1% growth supplement and 2% sodium chloride.

Violet Red Bile Agar with MUG is used for the detection of coliforms and the fluorogenic detection of Escherichia coli. Violet Red Bile Agar with MUG is not intended for use in the diagnosis of disease or other conditions in humans. Violet Red Bile Agar W/ MUG is specified in many procedures to enumerate coliforms in food and dairy products. In 1982, Feng and Hartman developed a rapid fluorogenic assay for Escherichia coli by incorporating 4-methylumbelliferyl-β-D-glucuronide (MUG) into Lauryl Tryptose Broth. Similarly, incorporating MUG into Violet Red Bile Agar permits the detection of E. coli among the coliform colonies. Standard methods procedures specify Violet Red Bile Agar with MUG for detecting E. coli in food and dairy products by fluorescence. This medium is often abbreviated as VRBA w/ MUG.

Reinforced Clostridial Agar can be used for the enumeration of anaerobes by pour plate, shake tube or membrane filtration methods, and is not intended for use in the diagnosis of disease or other conditions in humans. This is a solidified version of Reinforced Clostridial Medium (NCM0102) and can be used for the enumeration of anaerobes by pour plate, shake tube or membrane filtration methods. When solidified in tubes or bottles with minimal head space it can be used for anaerobic culture without the need for anaerobic atmosphere.

MacConkey Broth Purple is used for the detection and enumeration of fecal coliforms (37°C) and E. coli (44°C). This medium is not intended for use in the diagnosis of disease or other conditions in humans. The replacement of neutral red used in the original formulation by bromocresol purple makes the color change caused by acid producing organisms easier to read.
